A Histopathological Research About the Effects of Dimenhydrinate and Ondansetron were Used on Pregnant Rats on the Young Rats' Organs During Postnatal Period

ÖZET
Amaç: Gebe ratlarda kullanılan dimenhidrinat (DMH) ve ondansetronun postnatal dönemdeki yavru ratların organlarında oluşturacağı etkilerin histopatolojik yönden araştırılması amaçlandı. Gereç ve Yöntemler: Çalışmada 30 tane Wistar albino cinsi dişi rat kullanıldı. Ratlardan grup 1 (kontrol, n: 10), grup 2 (DMH, n: 10) ve grup 3 (ondansetron, n: 10) olmak üzere 3 grup oluşturuldu. Hamile kalmaları sağlandı ve hamile ratlara hamileliğin ilk gününden itibaren bir hafta süreyle 115 mg/kg/gün olmak üzere intramuskuler DMH (grup 2), 10 mg/kg/gün olmak üzere intraperitoneal ondansetron (grup 3) uygulandı. Kontrol grubuna (grup 1) ise aynı süre boyunca intramuskuler %0.9'luk NaCl verildi. Doğum sonrası yavru sayıları tespit edildi. Yavrulardan; yenidoğan, laktasyon ve erişkin dönemlerinde her gruptan beyin, akciğer, kalp, karaciğer ve böbrek dokuları alındı. Histolojik doku takip yöntemlerinden sonra preparatlar Hematoksilen-Eozin (H&E) ile boyanarak ışık mikroskobunda değerlendirildi. Bulgular: Kontrol grubu ile deney gruplarının karşılaştırılmasında yavru ratların organlarında histopatolojik olarak anlamlı bir değişiklik izlenmedi. Sonuç: Gebe ratlarda kullanılan DMH ve ondansetronun yavru ratların organlarında histopatolojik yönden herhangi bir değişikliğe neden olmadığı sonucuna varıldı.
ABSTRACT
Objective: We aimed to research the effects of Dimenhydrinate (DMH) and Ondansetron, used during pregnancy, on the organs of young rats in postnatal period in terms of histopathologically. Material and Methods: In the study, 30 Wistar-Albino type female rats have been used. The rats have been divided into three groups: group 1 (control, n: 10), group 2 (DMH, n: 10) and group 3 (ondansetron, n: 10). We kept them together for pregnancy starting from the first day of pregnancy, a dose of 115 mg/kg/day intramuscular DMH (group 2) and 10 mg/kg/day intraperitoneally ondansetron (group 3) have been given to pregnant rats for a week. As for the control group (group 1), a dose of intramuscular 0.9% NaCl has been given for the same period of time. After birth, the numbers of babies have been ascertained. Brain, lung, heart, liver and kidney tissues have been taken from the babies of each group during their newborn, lactation and adulthood periods. After histologic tissue follow-up methods, the preparates have been painted with Haematoxylin-Eosin (H&E) and then evaluated under light microscope. Results: There was no significant difference between the controls and experiment groups in point of histopathological change on the organs of the young rats. Conclusion: It has been concluded that DMH and ondansetron treatment during pregnancy period has no histopathological effect on the organs of the young rats.
